Wildcard Week



This week is about wildcard week.This week is very interesting because in this week we can make anything.We can make something creative using CNC machine.After wednesday's lecture our instructor suhas sir took brief lecture on same content.He told various interesting ideas for this week.Also my instructor Aditi shared various idea with me.

Individual Assignment: Design and produce something with a digital fabrication process (incorporating computer-aided design and manufacturing) not covered in another assignment,documenting the requirements that your assignment meets, and including everything necessary to reproduce it. Possibilities include (but are not limited to):

For this week I was thinking to make something new which was I never tried.In lecture Neil told about bioacademy.I searched about that and started thinking about what should I do.Until now I have worked in electronic design and mechanical design.Now its chance to work with biological term.I deciced to grow bacteria using one of cnc machine.In vigyan ashram we have microbiology lab.

I talked with Sonal ma'am who handles microbiology lab.She told me exact procedure and how to do it.She also told me which solutions are required.Also how to calculate percentage of solution which is required for me.

CNC Process:

For this assignment I used laser cutter for cnc process. I have made design of circular disc of my name in solidworks.It is simple design. I cut this design on acrylic sheet using laser cutter.It is required to grow bacteria on that plate.

After cnc process I went to microbiology lab.Sonal ma'am told me basic instructions of lab and I started actual work with the help of her .

Steps for bacteria plating are as follows:
1)Prepare media using autoclave.
2)Sterilize petri dish and beaker.
3)Pour media in petri dish.
4)Add bacteria which you want to grow.

Procedure:

Autoclave:For sterilization of media and glasswares.
Conditions:Temperature:121 degree celcius and pressure:15lbps

Laminar Air Flow(LAF):To maintain asceptic conditions.Keep it for minimum 10 min.(UV light)

Media:Provides nutrient for microorganism growth.
For fungus generally PDA (Potato Dextrose Agar)media is used and for bacteria NA media is used.
We are going to use PDA media .

Before starting with actual process I did calculation for PDA and Agar powder.For 1000ml of water they are taking 39gm of PDA ,so I have taken 300ml water and calculated PDA.Also I calculated for Agar powder.I have chosen Yeast as bacteria. I have taken 4gm of Yeast.I have added water in yeast .

Now we are started with actual work.Firstly I have taken PDA media as per calculation and also Agar powder which is used as solidifiyng agent.Then add distilled water in it.Shake it properly. Seal it with cotton plug.After this place media ,petri dish and acrylic dish in autoclave for sterilization. Keep this for 30 min.




This is the media which I prepared by adding PDA and Agar Powder.Figure shows the images of chemicals which I have used.




We decided to use Yeast as bacteria.We will grow bacteria in yeast.Also apart from this ,just for experiment we have added color in cutted part.Because of this we can see colorful bacteria.First we added media in petri dish and then I kept acrylic disc on it.Then we added color in cutted part ,that is I cut my name .I poured color on it. I kept it for two days.




After addition of color I added Yeast on it and make it same layer.Second picture shows the actual effect of color and bacteria added.



I checked it after two days and hopefully I got my result.Some amount of bacteria grown their.After seeing this I was very happy.Again I will check it after two days also I will check it under microscope.



Second Task:

After completion of first task I am thinking of doing something else.Then I decided to make foldable folder using laser cutter.For that I have used foldable pattern.I kept distance between two line was 2.76mm.I made design in coreldraw.I have taken butterfly image from internet and used trace bitmap command.




I cut this design on 3mm MDF material using laser cutter.I loved it .That butterfly looking very beautiful.

Learning outcome:

I was very happy about this week.I did something different and result of this also satisfying.I learned about technique of bacteria growing also different parameter which is related to this.Apart from this I learned how to make foldable design for laser cutter.
